Uncoupling of Spinach Thylakoids by Gramine

چکیده
Gramine and related alkaloids isolated from fodder plants in the Gramineae, Leguminoseae and other plant families [1] are responsible for the toxicity observed in ruminants grazing on these plants [2, 3]. Recently, these alkaloids were reported as toxic to aphids feeding on graminaceous crops [4]. Toxicity of gramine has been related to the known physiological effects of tryptamine related compounds [5]. However, a report on the effects of gramine on energy metabolism of mitochondria [6], provided an alternate mechanism to its toxic action. Further insight into the nature of these effects can be gained through a study of the effects of gramine on energy transduction in thylakoids. We herein report such study. This type of study has been used to assess the importance as allelochemicals of other indol and peptide alkaloids [7-9].

Effects of Allelochemical Gramine on Metabolic Activity and Ultrastructure of Cyanobacterium Microcystis aeruginosa
In this study, inhibition of Microcystis aeruginosa by antialgal alleochemical gramine, was studied by analyzing algal metabolic activity (represented by esterase and total dehydrogenase activities) and cell ultrastructure (showing morphological and ultrastructure alterations using transmission electron microscopy and DNA ladder analysis). After gramine exposure, esterase and total dehydrogenas...
متن کاملSulfhydryl reagents and energy-linked reactions in monocot thylakoids.
Monofunctional maleimides have been used to covalently modify the coupling factor protein of monocot thylakoid membranes. As with dicot thylakoids, incubation of the monocot thylakoids with maleimides in the light but not in the dark results in inhibition of both ATP synthesis and hydrolysis. In the dark, sites on the gamma and epsilon subunits of maize Zea mays coupling factor 1 are modified a...
متن کاملEffect of Light and Chilling Temperatures on Chilling-sensitive and Chilling-resistant Plants. Pretreatment of Cucumber and Spinach Thylakoids in Vivo and in Vitro.
The effects of chilling temperatures, in light or dark, on the isolated thylakoids and leaf discs of cucumber (Cucumis sativa L. "Marketer") and spinach (Spinacia oleracea L. "Bloomsdale") were studied. The pretreatment of isolated thylakoids and leaf discs at 4 C in the dark did not affect the phenazine methosulfate-dependent phosphorylation, proton uptake, osmotic response to sucrose, Ca(2+)-...
متن کاملQuality control of photosystem II: direct imaging of the changes in the thylakoid structure and distribution of FtsH proteases in spinach chloroplasts under light stress.
Under light stress, the reaction center-binding protein D1 of PSII is photo-oxidatively damaged and removed from PSII complexes by proteases located in the chloroplast. A protease considered to be responsible for degradation of the damaged D1 protein is the metalloprotease FtsH. We showed previously that the active hexameric FtsH protease is abundant at the grana margin and the grana end membra...
متن کاملPhotosynthetic light reactions in chemically fixed spinach thylakoids.
After proper fixation with glutaraldehyde, spinach thylakoids retain both photosystem I and photosystem II activity. The photochemical activity of fixed thylakoids is highly resistant to 1% triton. This indicates that fixed thylakoids may be useful starting material for isolation of functional components which can be related to the original structure of the membrane.
